Abstract:

The biochemical pathways involved in hepatic enzyme activation play a critical role in drug metabolism and development. Hepatic enzymes, primarily those located in the liver, are crucial for the biotransformation of pharmaceuticals, impacting their efficacy and safety. These enzymes participate in phase I and phase II reactions, including oxidation, reduction, hydrolysis, conjugation, and methylation. Understanding the intricate mechanisms that regulate hepatic enzyme activation is essential for optimizing drug design and developing personalized medicine strategies. Modulating enzyme activity can enhance drug bioavailability and minimize adverse effects, which is particularly relevant in diseases like cancer, cardiovascular disorders, and neurodegenerative conditions. Recent advances in biochemistry have revealed the genetic, epigenetic, and environmental factors influencing hepatic enzyme activation, presenting promising therapeutic opportunities. This review explores the biochemical pathways governing hepatic enzyme activation, emphasizing cytochrome P450 enzymes, phase II conjugating enzymes, and their interactions with signaling pathways. It also addresses challenges like interindividual variability in enzyme activity and the need for precision medicine approaches to improve therapeutic outcomes. Through better understanding of these pathways, researchers can predict drug interactions, optimize dosage regimens, and enhance therapeutic potential.
